Oncolytics Biotech® Receives Fast Track Designation for Pelareorep in Second-Line and Later Anal Cancer
July 20, 2026 09:00 ET

Designation supports development in an estimated $1 billion market where there are no FDA-approved therapies for patients whose disease progresses following first-line standard of care therapy

Third Fast Track designation in gastrointestinal cancers strengthens Oncolytics’ regulatory momentum and continues to advance the Company’s path toward registration

SAN DIEGO, July 20, 2026 (GLOBE NEWSWIRE) — Oncolytics Biotech® Inc. (Nasdaq: ONCY) (“Oncolytics” or the “Company”), a clinical-stage immunotherapy company, today announced that the U.S. Food and Drug Administration (“FDA”) has granted Fast Track designation to pelareorep in combination with a checkpoint inhibitor for the treatment of patients with inoperable, locally recurrent or metastatic squamous cell carcinoma of the anal canal (“SCAC”) who have progressed on or were intolerant to one or more prior lines of systemic therapy.

Fast Track designation is intended to facilitate the development and expedite the review of therapies that treat serious diseases and address significant unmet medical needs. The designation provides opportunities for more frequent interactions with the FDA throughout development, rolling review of a future Biologics License Application, and eligibility for Priority Review, if applicable.

The Company believes the designation further validates pelareorep’s potential to address a significant unmet need in advanced SCAC and follows the positive feedback received during its April 2026 meeting with the FDA regarding a potential registrational development strategy for the program. Based on the FDA’s feedback, Oncolytics believes it has a clear regulatory framework in place to advance pelareorep toward a pivotal study in this indication.

“This Fast Track designation represents another important regulatory milestone for Oncolytics and reflects the significant progress we have made over the past twelve months in advancing pelareorep toward potential registration,” said Jared Kelly, Chief Executive Officer of Oncolytics. “During that time, we have received Fast Track designation in both metastatic colorectal cancer and squamous cell anal carcinoma, achieved important FDA alignment on registrational development strategies, strengthened our intellectual property portfolio, and continued to generate compelling clinical data across our gastrointestinal oncology programs. Our April meeting with the FDA provided important clarity regarding a potential registrational pathway for pelareorep in second-line and later anal cancer, reinforcing our confidence in the program and our strategy to address an area of significant unmet need. We believe these milestones continue to de-risk our development programs while positioning the Company to deliver meaningful value for patients and shareholders.”

The designation is supported by encouraging efficacy demonstrated in the Company’s GOBLET study evaluating pelareorep in combination with a checkpoint inhibitor in patients with advanced SCAC whose disease had progressed following prior therapy. Previously reported clinical results demonstrated an objective response rate of approximately 30%, more than double historical response rates reported in this setting, a median duration of response of approximately 15.5 months versus 9.5 months, and 12-month overall survival rate of 82% compared to 45.7%.1, 2

With checkpoint inhibitor plus chemotherapy now established as the first-line standard of care in SCAC, there remains no FDA-approved therapies for patients whose disease progresses following such treatment, representing a significant unmet medical need and an estimated U.S. commercial opportunity approaching $1 billion annually.3, 4 Although checkpoint therapy is also approved as a single agent following progression or intolerance to platinum-based chemotherapy alone, no checkpoint inhibitor has been approved for patients who progress after receiving the current first-line standard of care regimen.

The Fast Track designation in SCAC is the third gastrointestinal cancer designation granted to pelareorep by the FDA, following designations in KRAS-mutated metastatic colorectal cancer in February 2026 and pancreatic cancer in late 2022. Together, these designations underscore pelareorep’s potential to address significant unmet medical needs and further validate the Company’s strategy of pursuing registration in high-value gastrointestinal cancer indications.

Half of planned clinical sites will be activated this month; more than 20 patients have been pre-identified across participating centers

Company to hold Type D meeting with FDA to discuss a potential registrational pathway through the addition of Part B to the existing REO 033 trial design

Part B will be a randomized study designed to support potential accelerated and full approval

SAN DIEGO, July 13, 2026 (GLOBE NEWSWIRE) — Oncolytics Biotech® Inc. (Nasdaq: ONCY) (“Oncolytics” or the “Company”), a clinical-stage company developing pelareorep, an investigational, systemically active immunotherapy that promotes potentially protective immune responses, including the upregulation of key inflammatory cytokines resulting in the formation of tertiary lymphoid structures and the expansion of tumor-infiltrating lymphocytes, today provided a clinical and regulatory update on REO 033, the Company’s randomized controlled study evaluating pelareorep in combination with folinic acid, fluorouracil and irinotecan (“FOLFIRI”) and bevacizumab for the second-line treatment of patients with Rat Sarcoma (“RAS”)-mutant, microsatellite stable (“MSS”) metastatic colorectal cancer.

REO 033 builds upon the previously reported REO 022 study, which more than doubled historical standard-of-care benchmarks across progression-free survival, overall survival, duration of response, and objective response rate.1-4 Based on these data, pelareorep has received Fast Track designation from the U.S. Food and Drug Administration (“FDA”) for this indication. The multi-part randomized REO 033 study is designed to prospectively validate these encouraging findings against a contemporary control arm while advancing pelareorep toward a potential registration pathway. The Company continues to make rapid operational progress in Part A of REO 033 (n=60 patients), with approximately half of the planned clinical sites activated by the end of July, and more than 20 patients have been pre-identified across participating centers. The remaining sites are expected to be activated by the end of August, positioning the study for accelerated enrollment during the second half of 2026. Most recently, global lead principal investigator Dr. Sanjay Goel and his team opened enrollment at Rutgers Cancer Institute of New Jersey.

“The magnitude and durability of the efficacy observed in REO 022 warrant earnest evaluation in a randomized setting,” said Dr. Sanjay Goel, Global Lead Principal Investigator for REO 033 and Professor of Medicine and Director of the Phase I Program at Rutgers Cancer Institute of New Jersey. “It is encouraging to see REO 033 expanding rapidly across leading academic centers, and I believe this study has the potential to further define the role of pelareorep in RAS-mutant MSS metastatic colorectal cancer.”

The Company also announced that it will hold a Type D meeting with the FDA in the first half of August 2026 to discuss the registrational design for REO 033 through the addition of Part B of the study. Building on the currently enrolling Part A of the study, this new registration-directed Part B would preserve the core design elements of REO 033 while increasing enrollment and incorporating blinded independent central review to support both a potential accelerated approval and a traditional full approval within the same study. The Company intends to align with the FDA on a registrational pathway that preserves the operational efficiencies already established through REO 033 while maintaining continuity with the existing clinical program under a prospectively agreed regulatory framework.

“Launching a global randomized study with multiple high-quality sites in a short period of time reflects the operational capabilities of our clinical organization and our investigators,” said Jared Kelly, Chief Executive Officer of Oncolytics. “Just as importantly, our ongoing interactions with the FDA have enabled us to focus on efficiently transitioning REO 033 into a registration-directed program built upon the existing trial infrastructure. We believe this strategy has the potential to significantly reduce development timelines while maintaining scientific rigor as we work to bring a much-needed immunotherapeutic option to patients in a treatment landscape that desperately needs innovation.”

The Company believes this approach provides the opportunity to generate early randomized efficacy data from Part A while simultaneously positioning Part B as a potential registrational study without the need to initiate a separate registrational trial. It also expects to report an initial tumor response update from patients enrolled in Part A by year-end 2026 and, subject to FDA feedback, initiate enrollment in Part B of the study during the first quarter of 2027.

About REO 033
REO 033 is a multi-part randomized controlled clinical trial evaluating pelareorep in combination with FOLFIRI and bevacizumab versus FOLFIRI and bevacizumab alone in patients with second-line RAS-mutant, microsatellite stable metastatic colorectal cancer (link to study on ClinicalTrials.gov). The study is designed to confirm the encouraging efficacy signals observed in REO 022 while generating the controlled clinical data necessary to support future regulatory interactions and potential registration.

Early results from solid tumor model support additional studies in pancreatic and colorectal cancer models; full dataset planned for presentation later in 2026

June 01, 2026 09:00 ET

SAN DIEGO, June 01, 2026 (GLOBE NEWSWIRE) — Oncolytics Biotech® Inc. (Nasdaq: ONCY) (“Oncolytics” or the “Company”), a clinical-stage company developing pelareorep, today announced initial data from a preclinical study evaluating pelareorep in combination with RAS inhibitor modalities in a solid tumor model, which demonstrate evidence of greater anti-tumor activity in combination than with the individual approaches alone. Pelareorep is an investigational, systemically active immunotherapy that promotes potentially protective immune responses, including the upregulation of key inflammatory cytokines resulting in the formation of tertiary lymphoid structures and the expansion of tumor-infiltrating lymphocytes.

Based on these findings, the Company is planning additional studies in models of pancreatic ductal adenocarcinoma (“PDAC”) and colorectal cancer (“CRC”) designed to further evaluate the combinations’ effects on immune activation, tumor response durability, and time-to-resistance. The ongoing work includes evaluations of pelareorep in combination with KRAS G12C inhibitors, pan-RAS inhibitors, and additional next-generation RAS pathway-targeting agents in RAS-mutated tumor models.

“We believe these initial findings further support pelareorep’s potential to serve as an immune-priming backbone for next-generation targeted therapies,” said Jared Kelly, Chief Executive Officer of Oncolytics. “RAS-mutated tumors, particularly pancreatic and colorectal cancers, remain among the most difficult cancers to treat due to intrinsic immune resistance and the emergence of therapeutic resistance over time. These preclinical results support further study of the combinations’ anti-tumor and immune effects, including in models designed to assess durability and time-to-resistance.”

Mr. Kelly continued, “Importantly, we believe this strategy may ultimately represent a potentially important area for further investigation in pancreatic cancer, where nearly all tumors harbor RAS pathway alterations and where patients continue to face extremely limited treatment options. The potential synergy observed in previous PDAC clinical studies and preclinical RAS-targeted modalities reinforces our belief that pelareorep may play an important role in future combination strategies designed to improve the durability of targeted therapies.”

Full results from the initial preclinical studies are expected to be presented in the fall or winter of 2026.

Translational data demonstrate induction of both innate and adaptive antiviral and anti-tumor specific immune responses capable of controlling tumor growth

May 22, 2026 09:00 ET

SAN DIEGO, May 22, 2026 (GLOBE NEWSWIRE) — Oncolytics Biotech® Inc. (Nasdaq: ONCY) (“Oncolytics” or the “Company”), a clinical-stage company developing pelareorep, today announced translational data from the GOBLET and AWARE-1 trials will be presented at the 2026 American Society of Clinical Oncology (“ASCO”) Annual Meeting at McCormick Place, Chicago, Illinois, from May 29-June 2, 2026. Pelareorep is an investigational, systemically active immunotherapy that promotes potentially protective immune responses, including the upregulation of key inflammatory cytokines resulting in the formation of tertiary lymphoid structures and the expansion of tumor-infiltrating lymphocytes.

“These findings demonstrate that pelareorep works by expanding preexisting tumor-reactive immune cells rather than introducing new antigens,” said Dr. Richard Vile, Ph.D., Professor of Immunology at the Mayo Clinic and Oncolytics Scientific Advisory Board Member. “This further underscores Oncolytics’ continued commitment to define the mechanisms by which pelareorep engages the immune system and illustrates the progress being made in developing a differentiated immunotherapy to improve treatment options for difficult-to-treat cancers.”

“KRAS-mutant tumors remain among the most challenging to treat with immunotherapy,” said Jared Kelly, Chief Executive Officer of Oncolytics. “These data reflect important progress in bridging translational and clinical data and help us better understand how immune responses are activated by pelareorep. When we can point to translational and mechanistic data as the reason for clinical efficacy, it gives us confidence that our development plans are more likely to succeed.”

Abstract: Influence of pelareorep on mutant KRAS-specific blood TIL clonal expansion.
Author: Richard Trauger, PhD
Date: May 30, 2026, 1:30-4:30 p.m. Central Time
Abstract Number: 2664
Poster Board Number: 454

Translational analyses from the Phase 1/2 GOBLET study in advanced pancreatic cancer and the AWARE-1 window-of-opportunity study in early-stage breast cancer illustrate a multi‑step immune activation process following treatment with pelareorep. Results demonstrate that pelareorep induces an antiviral immune response within the tumor via double-stranded RNA signaling, leading to expansion of virus‑specific T cells and subsequent activation of tumor‑specific T cells associated with tumor regression.

In pancreatic cancer, serial blood analyses demonstrated expansion of anti‑viral T cells by ELISPOT and clonal expansion of tumor‑specific T cells after one cycle of therapy. Notably, the expansion of pre‑existing tumor‑infiltrating lymphocyte (“TIL”) clones in blood correlated with reductions in tumor volume in pancreatic cancer. T‑cell receptor sequencing with antigen specificity assessment confirmed expansion of mutant KRAS-specific T‑cell clones, supporting a model in which pelareorep engages both innate and adaptive immunity and may help drive tumor‑directed immune responses in difficult‑to‑treat, KRAS‑driven disease.

In serial breast tumor biopsies, gene expression profiling demonstrated significant increases in antiviral and immune gene expression, consistent with activation of toll‑like receptor 3 (TLR3), alongside induction of CXCL13, a chemokine linked to the formation of tertiary lymphoid structures (TLS). TLS formation in the tumor microenvironment was further supported by imaging mass cytometry following treatment. Across studies, evidence of the expansion of TILs was observed in the tumor and the blood.

A copy of the ASCO presentation will be available on the Media page of the Oncolytics’ website following the conclusion of the meeting.

About AWARE-1
AWARE-1 was an open-label window-of-opportunity study in early-stage breast cancer. The study combined pelareorep, without or with atezolizumab, and standard of care therapy according to breast cancer subtype. Tumor tissue was collected from patients as part of their initial breast cancer diagnosis, again on day three following initial treatment, and finally at three weeks following treatment, on the day their tumor is surgically resected. Key objectives of the study were to confirm that pelareorep is acting as a novel immunotherapy, to evaluate potential synergy between pelareorep and checkpoint blockade, and to collect biomarker data. The primary endpoint of the translational study was overall CelTIL score (a measurement of cellularity and tumor-infiltrating lymphocytes that is associated with favorable clinical outcomes). Secondary endpoints for the study included safety and tumor and blood-based biomarkers. The combination of pelareorep, letrozole, and atezolizumab resulted in 60% of patients experiencing 30% or greater increases in their CelTIL score.

2L KRAS-Mutant MSS mCRC Demonstrates ~3–4x Improvement in Duration of Response vs. Historical 4–6 Month Benchmark

Company Engaged with FDA to Support Potential Accelerated Approval Pathway in 2L RAS-Mutant MSS mCRC

SAN DIEGO, May 04, 2026 (GLOBE NEWSWIRE) — Oncolytics Biotech® Inc. (Nasdaq: ONCY) (“Oncolytics” or the “Company”), a clinical-stage company developing pelareorep, an investigational, systemically delivered immunotherapy that has been shown to activate innate immune-sensing pathways, today announced new durability data in metastatic colorectal cancer (“mCRC”), demonstrating meaningful and sustained clinical benefit in patients with RAS-mutant, microsatellite-stable (“MSS”) disease.

Pelareorep-based combination therapy demonstrated a 19.5-month median duration of response in second-line (“2L”) KRAS-mutant MSS mCRC patients in the REO 022 study, compared to historical benchmarks of approximately 4–6 months in this setting.1 Additional data from this study include an objective response rate of 33% for patients receiving pelareorep, bevacizumab, and FOLFIRI, tripling the 6-11% for the standard of care.2, 3

“We believe these data demonstrate a compelling durability signal for pelareorep in colorectal cancer,” said Jared Kelly, Chief Executive Officer of Oncolytics. “A 19.5-month median duration of response in second-line patients—representing a three- to four-fold improvement over historical expectations—highlights pelareorep’s potential to deliver sustained benefit in a population with few effective options. We believe these results support a path toward accelerated approval in second-line RAS-mutant MSS metastatic colorectal cancer, and we are actively engaging with the FDA to align on a regulatory strategy leveraging our ongoing randomized study.”

Oncolytics is currently enrolling patients in its randomized Phase 2 study evaluating pelareorep in combination with FOLFIRI and bevacizumab in second-line RAS-mutant MSS mCRC (link to study on ClinicalTrials.gov). The Company is actively engaging with the U.S. Food and Drug Administration (“FDA”) to discuss a potential accelerated approval pathway based on response durability and time-to-event endpoints from this study.

Colorectal cancer remains one of the largest oncology markets globally, with significant unmet need in later-line settings. RAS-mutant MSS mCRC represents a particularly difficult-to-treat population, where patients typically experience rapid disease progression and limited durability of response on standard therapies. The magnitude and consistency of durability and patient response observed with pelareorep-based combinations suggest the potential to meaningfully extend clinical benefit in this setting.

About Oncolytics Biotech

Innately Adaptive™

Oncolytics Biotech Inc. is developing pelareorep, a safe and well-tolerated intravenously delivered immuno-oncolytic virus (IOV) that kills cancer through a unique mechanism of action with two components, selective tumor lysis and activation of the innate and adaptive immune systems, creating an inflamed phenotype to treat a variety of solid tumors and hematological malignancies.

Developing Potential Treatments for a Variety of Tumor Types

Oncolytics Biotech Inc. is developing an intravenously delivered immuno-oncolytic virus called pelareorep, for the treatment of solid tumors and hematological malignancies. Pelareorep is a non-pathogenic, proprietary isolate of the unmodified reovirus that: 1) induces selective tumor lysis, and 2) promotes an inflamed tumor phenotype through innate and adaptive immune responses. Our clinical development plan is based on these two components of the mechanism of action, with a focus on obtaining regulatory approval as soon as possible.

Partnering, Collaborating & Combining Towards the Future

Oncolytics will grow through partnerships, collaborations and combinations, including:

  • Co-development agreement with Pfizer & Merck KGaA to study their PD-L1 checkpoint inhibitor Bavencio® in combination with pelareorep in HR+ /HER2- metastatic breast cancer.
  • Master Supply Agreement with Roche to supply Tecentriq® – their PD-L1 checkpoint inhibitor – for Oncolytics’ clinical development program. This was put to use immediately in the AWARE-1 study in breast cancer, a collaboration with the cooperative group SOLTI
  • Combination studies with Merck’s PD-1 checkpoint inhibitor Keytruda® in both multiple myeloma and pancreatic cancer.
  • Combination study with Bristol-Myers Squibb’s PD-1 checkpoint inhibitor Opdivo® in multiple myeloma.
  • Partnership with Adlai Nortye in China to develop pelareorep for metastatic breast cancer, as well as potentially additional indications

We continue to investigate further collaboration and partnership opportunities, particularly with checkpoint inhibitors and other immuno-oncology drugs, that will help us to advance pelareorep in the adjuvant setting.

Manufacturing

Pelareorep is currently being manufactured at commercial scale in its final formulation under a commercial supply agreement with MilliporeSigma.

Clinical Development Plan

Oncolytics is currently focused on immunotherapy combinations, including our studies with Bavencio®, Keytruda®, Opdivo® and Tecentriq®.

Our clinical development plan is based on pelareorep’s broad applicability to boost the effectiveness of a range of immunotherapies – with an immediate focus on checkpoint inhibitors – across multiple cancer indications. The primary objective is to obtain regulatory approval for pelareorep as quickly as possible and is based on our compelling metastatic breast cancer survival data, our synergies with checkpoint approved inhibitors and our biomarker. The second objective is to expand pelareorep into commercially valuable new treatment areas including other immunotherapy agents in collaboration with pharmaceutical partners.

We are excited to work with Pfizer, Merck KGaA, Merck, Bristol-Myers Squibb and Roche with their checkpoint inhibitors, and look forward to developing our programs to evaluate the efficacy and safety in our clinical studies targeting metastatic breast cancer, early stage breast cancer, multiple myeloma and pancreatic cancer.   There is great potential to eventually work with additional immunotherapies in additional indications and this will only drive the potential for treatment with pelareorep.

Collaborators

Based on the Company’s Clinical Development Plan, Oncolytics is investigating potential partnerships for its phase 3 registration program in metastatic breast cancer (mBC), as well as additional collaborations with checkpoint inhibitors that will help advance pelareorep in different indications and disease states, including the adjuvant setting.

Partnering

We are working with Adlai Nortye, a biopharmaceutical company focused on discovering and developing important new treatments for cancer and metabolic diseases, to advance our phase 3 registration program in mBC and to leverage their distinct experience and expertise in drug development and commercialization in China, Hong Kong, Macau, Singapore, South Korea and Taiwan. Under the terms of the agreement, Adlai Nortye has exclusive development and commercialization rights to pelareorep for all indications in these territories.

Co-Development

Oncolytics entered into a co-development agreement with Pfizer and Merck KGaA to develop pelareorep in combination with paclitaxel and Bavencio®, a human anti-PD-L1 antibody, for the treatment of hormone-receptor positive, human epidermal growth factor 2-negative (HR+ / HER2-) metastatic breast cancer. Oncolytics and Pfizer will share costs associated with the phase 2 clinical trial called BRACELET-1 (BReast cAnCEr with the Oncolytic Reovirus PeLareorEp in CombinaTion with anti-PD-L1 and Paclitaxel), an open label study that will enroll 45 patients into three cohorts with 15 patients per cohort: paclitaxel alone, paclitaxel in combination with pelareorep and paclitaxel in combination with both pelareorep and avelumab. The study will examine the expression of immune-related biomarkers to identify changes in T cell clonality between pre-treatment and on-therapy biopsies to confirm our previously identified biomarker and is designed to assess efficacy in terms of overall response rate at week 16 per RECIST 1.1 and iRECIST. The safety of the combination will also be evaluated.

Collaborations & Supply Agreements

Solti

The Window of Opportunity (WOO) study in breast cancer, in collaboration with SOLTI, an academic research group dedicated to clinical and translational research in breast cancer, is being sponsored by Oncolytics and facilitated by SOLTI. The WOO study is in the neoadjuvant setting for breast cancer where patients will receive the appropriate standard of care for their cancer subtype plus pelareorep with or without the anti-PD-L1 cancer immunotherapy atezolizumab (Tecentriq®). The study will be coordinated by Dr. Aleix Prat, Head of Medical Oncology at the Hospital Clínic of Barcelona, Associate Professor of the University of Barcelona and the Head of the Translational Genomics and Targeted Therapeutics in Solid Tumors Group at August Pi i Sunyer Biomedical Research Institute (IDIBAPS) and member of Oncolytics’ Scientific Advisory Board. SOLTI has a network of more than 300 professionals, mostly medical oncologists, in over 80 hospitals in Spain, Portugal, France and Italy. Final study design and other details will be announced upon enrollment of the first patient, expected early 2019.

Roche

Oncolytics entered into a Master Clinical Supply Agreement with Roche to supply atezolizumab (Tecentriq®) for use in the company’s clinical development program. The supply agreement enables Oncolytics to further investigate pelareorep’s impact on cancer treatments in combination with atezolizumab and has already been incorporated into the company’s clinical program via the SOLTI WOO study in breast cancer. Data from studies with Tecentriq will broaden Oncolytics’ experience with the entire drug class as the company looks to demonstrate the impact of pelareorep with checkpoint inhibitors. Under this five-year Master Clinical Supply Agreement, Roche will supply Tecentriq for the proposed clinical trial with both parties having access to the clinical data.

Combinations

Merck

Oncolytics is combining pelareorep with Merck’s anti-PD-1, pembrolizumab (Keytruda®), in two Investigator Sponsored Trials (IST’s). The goal is to expand the use of checkpoint inhibitors as anti-cancer agents by promoting an inflamed phenotype in the tumor or turning cold tumors hot.

The first study is in second line pancreatic cancer run by Dr. Devalingham Mahalingam. It will enroll approximately 40 patients with advanced pancreatic cancer and be conducted at the Robert H. Lurie Comprehensive Cancer Center of Northwestern University.

The second study, in collaboration with the Keck School of Medicine of University of Southern California (USC), is in multiple myeloma. This study, facilitated by Dr. Kevin Kelly, Associate Professor of Clinical Medicine, continues to build on the Company’s I-O program by combining proteasome inhibitors and pelareorep to demonstrate an increase of PD-L1 expression on multiple myeloma cells and recruitment of T-cells to the bone marrow.

Bristol-Myers Squibb

Oncolytics is combining pelareorep and nivolumab (Opdivo®) with a proteasome inhibitor for patients with relapsed/refractory multiple myeloma. The study will be facilitated by Craig C. Hofmeister, MD, MPH at Emory University and Douglas Sborov MD, MS at the University of Utah and expands on Oncolytics’ strategy of investigating the importance of systemic pelareorep administration followed by checkpoint blockade. This combination study will not only help demonstrate the specific role of pelareorep in promoting an inflamed phenotype, it should also help demonstrate how the virus can promote responses to checkpoint blockade in cold tumors.
